Totally possible:

Manually Mode - YES
It's simple:
START MENU -> CLOSE -> SLEEP
Note: I ask again how to undo your command (Restore default settings PowerCfg)?

Can not disable the sleep drive or computer in various method, eg. Commands or Windows (Power Options) or ASUS applications or application Hard Disk Sentinel because it depends on the application Intel Rapid Technology.

Link power management
Link power management (LPM) is an energy-saving technique that helps SATA devices to save energy. Speed SATA device is set to the low power during idle time when no active I / O device or are missing or not used. The LPM automatically sets the SATA link to an active state power when the I / O requests are queued to that link and tasks are to be performed.

The LPM is enabled by default for the controller and each connected device, the following requirements are met:
■ SATA connection must be enabled in the BIOS to manage energy consumption.
■ The system is restarted after joining SATA devices
■ Configuration Management platform takes into account the function of the power link
■ SSDs and hard drives support applications HIPM (Host-Initiated Power Management) and DIPM (Device-Initiated Link Power Management)

To enable the management of power connection, do the following:
1. Click on the 'Performance' and then click 'Settings efficiency and power' in the left pane.
2. In the area of power management link click 'Activate'.
3. For the changes to take effect, restart your computer.

Turn off power management link
1. Click on the 'Performance' and then click 'Settings efficiency and power' in the left pane.
2. In the area of power management link click 'Disable'.
