I have MSE, SuperantiSpyware (paid), and Malwarebytes(paid). I've been running full system scans (at different times of course) once a week for all three. Is this required or overkill?

Overkill
For me I rarely scan. I only do when something seems wrong. Haven't had a virus in over 6 years.
The real trick to keeping your self safe is keeping java and flash up to date. That is how a lot of infections get in now. Through security wholes in java and flash.
Every infection I have cleaned up on customers machines over the past 6 months have had outdated java and flash. Once updated they don't get hit again. Until a new whole is found of course
